ABSTRACT

The research evaluated the interactions of two main factors (strain / types of spawn) on various parameters with the purpose to assess its effect on yield and biochemical composition of Lentinula edodes fruiting bodies cultivated on pasteurized wheat straw. The evaluation was made with four strains (IE-40, IE-105, IE-124 and IE-256). Different types of spawns were prepared: Control (C) (millet seed, 100%), F1 (millet seed, 88.5%; wheat bran, 8.8%; peat moss, 1.3%; and CaS0(4), 1.3%) and F2 (the same formula as F1, but substituting the wheat bran with powdered wheat straw). Wheat straw was pasteurized by soaking it for 1 h in water heated to 65 °C. After this the substrate (2 kg wet weight) was placed in polypropylene bags. The bags were inoculated with each spawn (5% w/w) and incubated in a dark room at 25 °C. A proximate analysis of mature fruiting bodies was conducted. The mean Biological Efficiency (BE) varied between 66.0% (C-IE-256) and 320.1% (F1-IE-124), with an average per strain of 125.6%. The highest mean BE was observed on spawn F1 (188.3%), significantly different from C and F2. The protein content of fruiting bodies was high, particularly in strain IE-40-F1 (17.7%). The amount of fat varied from 1.1 (F1-IE-40) to 2.1% (F2-IE-105) on dry matter. Carbohydrates ranged from 58.8% (F1-IE-40) to 66.1% (F1-IE-256). The energy value determined ranged from 302.9 kcal (F1-IE-40) to 332.0 kcal (F1-IE-256). The variability on BE observed in this study was significantly influenced by the spawn's formulation and genetic factors of the different strains.

ABSTRACT

Como parte de una posible producción de basidiocarpos de Pleurotus eryngii en Chile, se seleccionó como sustrato paja de trigo. Como objetivo principal se evaluó la eficiencia de dos tratamientos térmicos, para reducir la carga microbiana del sustrato, además de determinar los cambios en la relación C:N antes y después a los tratamientos térmicos y cosechas de basidiocarpos. Al mismo tiempo, se determinó la eficiencia biológica de P. eryngii. En un primer tratamiento, partidas del sustrato fueron sometidas a ebullición directa por 1 h., y en un segundo se usó la pasteurización por 3 horas. El sustrato de ambos tratamientos se depositó en bolsas de nylon de 8 Kg y se sembraron con ®semilla¼ de P. eryngii y se incubaron hasta la obtención de basidiocarpos. Se determinó en el sustrato el C y N según Saavedra (1975), y la celulosa, hemicelulosa, lignina y extraíbles totales según métodos propuestos por las normas Tappi (2000). La mayor eficiencia biológica con P. eryngii, se logró en el sustrato sometido a pasteurización.


In an attempt to get a possible production of Pleurotus eryngii basidiocarps, wheat straw was selected as a substratum. The main purpose was to assess the efficiency of two thermal treatments in order to reduce the microbial load of the substratum as well as to detect any change in the C:N relation prior and after thermal treatments and basidiocarp harvests. Besides the biological efficiency of P.eryngii was determined. In a first treatment, some parties of the substratum were submitted to direct boiling for 1h. while in a second one, pasteurization was used for 3h. The resulting substrata of both treatments were kept in 8-kg nylon bags and then sowed with P.eryngii ®seed¼and later on incubated in order to get basidiocarps. C and N were determined in the substratum according to Saavedra (1975) while cellulose, hemicellulose, lignin and removable totals were determined following the methods proposed by Tappi standards (2000). The substratrum submitted to pasteurization revealed the highest biological efficiency with P.eryngii.

ABSTRACT

Cellulolytic properties of two white rot fungi, Bjerkandera adusta and Pycnoporus sanguineus, cultivated on wheat straw agar medium, were characterized and compared. Optimal growing parameters for maximum enzyme production for both fungi were wheat straw medium pH 5 and 28ºC. B. adusta showed, on the 6th day of culture, carboxymethylcellulose (CMC)ase activity levels 1.6 times higher than maximal P. sanguineus activity, achieved on the 8th day. B. adusta supernatants also displayed higher activity levels towards xylan (3.6-fold) compared to those of P. sanguineus. However, enzymes from P. sanguineus were more robust resisting one hour incubation at high temperatures (up to 80ºC), and exhibiting activity and stability in pH range from 2 to 8. Cellulolytic activities, with molecular masses ranging from 25 to 90 kDa, from the two species were detected in zymograms.

ABSTRACT

The effect of Pleurotus pulmonarius on the chemical composition of wheat straw was evaluated. Wheat straw, treated and untreated with P. pulmonarius, was obtained from a commercial facility. Ten samples plastic bags of wheat straw used previously as substrate to culture edible fungus were collected at random. The negative control group consisted of the pasteurized wheat straw untreated with P. pulmonarius. All samples were analyzed to determine dry matter, organic matter, crude protein, neutral detergent fiber, acid detergent fiber, cellulose and hemicellulose of each wheat straw. Data were analyzed by mean comparison using a t-Student test. No differences (P>0.05) between treatments were found for dry matter, crude protein and hemicellulose; however, straw treated with P. pulmonarius showed higher percentages (P<0.05) of organic matter, neutral and acid detergent fiber. It is concluded that growing P. pulmonarius in wheat straw improves the chemical composition of the straw by increasing its organic matter content and modifies the fiber structure, which increases the soluble carbohydrates content.


Se midió el efecto en la composición química de la paja de trigo usada como sustrato en la producción de Pleurotus pulmonarius. Las pajas de trigo, tanto tratadas con P. pulmonarius como no tratadas, fueron obtenidas de una empresa comercial. Para obtener muestras representativas para el análisis químico se tomaron al azar diez bolsas de la paja usada como sustrato para cultivar el hongo. El grupo testigo consistió en las pajas esterilizadas sin tratamiento con P. pulmonarius. A todas las muestras se les determinó materia seca, materia orgánica, proteína cruda, fibra detergente neutro, fibra ácido detergente, celulosa y hemicelulosa. Los datos obtenidos fueron analizados mediante una comparación de medias utilizando una prueba de t-Student. No se encontraron diferencias (P>0,05) para el porcentaje de materia seca, proteína cruda y hemicelulosa; sin embargo, se presentó un mayor (P<0,05) porcentaje de materia orgánica, fibra detergente neutro y ácido para la paja tratada en comparación con la no tratada. Se concluye que el crecimiento del P. pulmonarius en paja de trigo mejora la composición química de las pajas aumentando el contenido de materia orgánica y modifica la estructura de la fibra, con lo que se incrementa el contenido de carbohidratos solubles.


Mede-se o efeito na composição química da palha de trigo usada como substrato na produção de Pleurotus pulmonarius. As palhas de trigo, tanto tratadas com P. pulmonarius como não tratadas, foram obtidas de uma empresa comercial. Para obter amostras representativas para a análise química se tomaram aleatoriamente dez sacolas da palha usada como substrato para cultivar o fungo. O grupo testemunha consistiu em palhas esterilizadas sem tratamento com P. pulmonarius. Determinou-se, em todas as amostras, matéria seca, matéria orgânica, proteína crua, fibra detergente neutro, fibra ácido detergente, celulose e hemicelulose. Os dados obtidos foram analisados mediante uma comparação de médias utilizando uma prova de t. Não se encontraram diferenças (P>0,05) para a porcentagem de matéria seca, proteína crua e hemicelulose; no entanto, se apresentou uma maior (P<0,05) porcentagem de matéria orgânica, fibra detergente neutra e ácida para a palha tratada em comparação com a não tratada. Conclui-se que o crescimento do P. pulmonarius na palha de trigo melhora a composição química das palhas aumentando o conteúdo de matéria orgânica e modifica a estrutura da fibra, com o que se incrementa o conteúdo de carboidratos solúveis.

ABSTRACT

Wheat straw hemicellulosic hydrolysate was used for xylitol bioproduction. The use of a xylose-containing medium to grow the inoculum did not favor the production of xylitol in the hydrolysate, which was submitted to a previous detoxification treatment with 2.5 percent activated charcoal for optimized removal of inhibitory compounds.


Hidrolisado hemicelulósico de palha de trigo foi utilizado para a bioprodução de xilitol. O uso de meio contendo xilose para crescer o inóculo não favoreceu a produção de xilitol no hidrolisado, que foi submetido a um tratamento prévio de destoxificação com 2.5 por cento de carvão ativo para remoção otimizada de compostos inibitórios.

ABSTRACT

A new thermophilic fungus J18 isolated from the soil samples was identified as Paecilomyces thermophila. This strain produced effectively xylanase utilizing several lignocellulosic materials in the solid-state fermentation (SSF) , and wheat straw was the best carbon source. The results of single-factor-experiment showed that the wheat straw of particle size 0. 3 mm ~ 0.45 mm, initial moisture content of 83% , initial pH of 7. 0 and cultivation temperature of 50℃were the optimal conditions for xylanase production. Under the optimized conditions, it produced 18 580 U/g dry substrate after 8 days of cultivation. Therefore, xylanase production by Paecilomyces thermophila in SSF possess great potential for commercial applications.
